建立了一种电化学方法用于分析细胞凋亡和抗肿瘤药物的作用。应用特殊而简便的电极系统分析足叶乙甙(Vp-16)诱导的K562细胞凋亡。观察凋亡细胞的形态学改变和被动电化学行为。结果表明:DNA凝胶电泳和流式细胞仪分析均显示出典型的细胞凋亡特征,同时凋亡细胞的电化学伏安行为也明显不同,表现为凋亡细胞的峰值电流和电子转移速率的下降。结论提示电化学方法不仅能够反映细胞凋亡,尤其是早期细胞凋亡,而且提供了一种评价活细胞氧化还原系统改变的方法。
An electrochemical method was developed to analyze the role of apoptosis and antineoplastic agents. The apoptosis of K562 cells induced by etoposide (Vp-16) was analyzed using a special and simple electrode system. Morphological changes and passive electrochemical behaviors of apoptotic cells were observed. The results showed that both DNA gel electrophoresis and flow cytometry analysis showed the typical characteristics of apoptosis. At the same time, the electrochemical voltammetric behavior of apoptotic cells was also significantly different, showing the peak current and electron transfer rate of apoptotic cells decline. The results suggest that electrochemical methods can not only reflect apoptosis, especially early apoptosis, but also provide a method to evaluate the change of redox system in living cells.
