目的研究葛根粗提物和葛根素对乙醇引起的胎鼠海马细胞氧化损伤是否具有相同的保护作用。方法海马细胞的培养取材于孕18d的ICR胎鼠,采用RT-PCR和Western印迹方法观察不同剂量的葛根粗提物及葛根素对乙醇引起的胎鼠海马细胞HSP70mRNA及蛋白质表达的变化。结果15mg/L葛根粗提物和10mg/L葛根素均可部分拮抗50~350mmol/L不同浓度乙醇引起的胎鼠海马细胞HSP70表达量的升高。结论葛根粗提物和葛根素均可降低乙醇所致海马细胞HSP70表达量的升高,证实两者具有相同的抗氧化作用。
OBJECTIVE: To study whether pueraria crude extract and puerarin have the same protective effect on ethanol-induced oxidative damage of fetal rat hippocampal cells. Methods The cultured hippocampal cells were obtained from ICR fetuses at the 18th day of pregnancy. RT-PCR and Western blotting methods were used to observe the changes of HSP70 mRNA and protein expression in ethanol-induced fetal rat hippocampal cells with different doses of crude extracts of Radix Puerariae and puerarin. Results The crude extracts of Pueraria lobata at 15 mg/L and 10 mg/L puerarin could partially antagonize the increase of HSP70 expression in fetal rat hippocampus induced by 50-350 mmol/L ethanol. Conclusion Pueraria lobata extract and puerarin can reduce the increase of HSP70 expression in hippocampal cells induced by ethanol, confirming that both have the same antioxidant effect.
