目的W基因是利用周围神经髓鞘蛋白PMP-22基因的编码区5′-及3′-序列为引物,藉PCR方法自人脑胶质瘤cDNA文库中扩增的一个基因片段,本实验研究W基因组织表达的特异性。方法搜集不同组织来源的肿瘤及正常脑组织标本,行RT-PCR及RNA斑点杂交检测。结果在非神经组织来源的肿瘤及正常脑组织中未检出W基因表达,在一例星形胶质瘤Ⅲ级组织可检测到W基因表达。RT-PCR及RNA斑点杂交结果一致。结论提示W基因表达可能具有组织特异性。
Objective W gene is a gene fragment amplified from the cDNA library of human glioma by PCR using the 5’-and 3’-sequences of the coding region of PMP-22 gene as a primer. The present study Specificity of W gene expression. Methods Tumors and normal brain tissue samples from different tissues were collected and detected by RT-PCR and RNA dot blot. Results W gene expression was not found in tumors and normal brain tissues of non-nerve tissue origin. W gene expression was detected in grade III astrocytoma. RT-PCR and RNA dot blot hybridization results. Conclusions suggest that W gene expression may be tissue-specific.
