目的 :研究益智复方对痴呆小鼠学习记忆及自由基代谢的影响。方法 :小鼠每日颈背部皮下注射D 半乳糖 0 .15 0g·kg-1,连续 4 5d可以造成痴呆模型。在注射半乳糖 5d后 ,同时灌胃给予不同剂量的YZC至造模结束 ,再采用Y型迷宫测验及SOD、MDA检验试剂盒分别查看其对痴呆小鼠学习记忆及自由基代谢的影响。结果 :益智复方可以显著提高痴呆小鼠Y型迷宫学习 记忆成绩 (所需的训练次数由模型组的 31.35± 8.97 17.12± 5 .5 7降低到低中高三个剂量组的 19.67± 8.0 7 11.2 7± 6.65、14.0 0± 6.84 8.2 9± 5 .95和 11.4 1± 5 .99 6.2 4±4 .97) ;益智复方 0 .10 8g·kg-1和 0 .4 33g·kg-1均可显著提高小鼠脑组织SOD活性 ,从 17.69± 3.14nU·mg-1pro升高到 2 6.94± 4 .4 6nU·mg-1pro和2 1.33± 3.63nU·mg-1pro ;而益智复方 0 .10 8g·kg-1、0 .2 17g·kg-1和 0 .4 33g·kg-1使小鼠大脑中丙二醛 (MDA)水平由 4 .0 8± 0 .88nmol·mg-1pro减少到 2 .82± 0 .75nmol·mg-1pro、 2 .10± 0 .4 2nmol·mg-1pro和 2 .35± 0 .39nmol·mg-1pro。结论 :益智复方具有较好的改善学习记忆功能 ,这可能与其清除脑内自由基、抑制脂质过氧化的作用有联系。
Objective: To study the effects of Yizhifu Recipe on learning and memory and free radical metabolism in mice with dementia. METHODS: Mice were injected subcutaneously with D-galactose at a dose of 0.15 kg · kg -1 every day for 4 days and 5 days. After injection of galactose for 5 days, different doses of YZC were intragastrically given to the end of modeling. Y-maze test and SOD, MDA test kit were used to examine the effects of YZC on learning and memory and free radical metabolism in mice with dementia respectively. Results: Yizhi compound could significantly improve learning and memory ability of Y maze in dementia mice (the number of training required was reduced from 31.35 ± 8.97 17.12 ± 5. 57 in model group to 19.67 ± 8.0 7 in low, medium and high dose groups 7 ± 6.65, 14.0 ± 6.84 8.2 9 ± 5.95, and 11.4 1 ± 5 .99 6.2 4 ± 4 .97), respectively; 0.81 g · kg-1 and 0.433 g · kg-1 Significantly increased SOD activity in mouse brain tissue from 17.69 ± 3.14nU · mg-1pro to 2 6.94 ± 4.64nU · mg-1pro and 2 1.33 ± 3.63nU · mg-1pro; The level of malondialdehyde (MDA) in the brain decreased from 4.08 ± 0.88nmol · mg-1pro at 10 8g · kg -1, 2.02g · kg -1 and 0.433g · kg -1 To 2.82 ± 0.75nmol · mg-1pro, 2.10 ± 0.42nmol · mg-1pro and 2.35 ± 0.39nmol · mg-1pro. Conclusion: Yizhi compound has good learning and memory abilities, which may be related to its function of clearing free radicals in the brain and inhibiting lipid peroxidation.
