目的:探讨喉鳞状细胞癌组织中缺氧诱导因子-1α(HIF-1α)、环氧合酶-2(COX-2)蛋白质和mRNA的表达,以期有助于喉鳞状细胞癌发生、发展和转移机制的进一步阐明。方法:用免疫组织化学和RT-PCR方法检测76例喉鳞状细胞癌组织和9例癌旁正常组织中HIF-1α、COX-2蛋白和mRNA的表达,并按T分期、病理分级分别探讨HIF-1α、COX-2蛋白和mRNA的表达与各临床病理参数的关系。结果:①HIF-1α、COX-2蛋白表达在喉癌标本的表达量明显高于癌旁正常组织(P<0.05),随T分期的增加而增高;中、低分化的喉癌组织中HIF-1α表达显著高于高分化喉癌;COX-2蛋白表达与病理分级无关。②HIF-1α与COX-2mRNA在喉癌组织中的阳性表达率分别是67.11%和75.00%,明显高于癌旁正常组织(P<0.05),癌旁正常组织未检测到COX-2mRNA表达。HIF-1α与COX-2mRNA在喉癌组织中的阳性表达率与T分期有关(P<0.05),标本的表达量随T分期的增加而增高,但是与病理分级无关(P>0.05)。③HIF-1α蛋白表达阳性病例中,COX-2的阳性率为78.72%。结论:HIF-1α与COX-2在喉鳞状细胞癌组织的表达增高并密切相关,可能是喉癌发生发展的原因之一。
Objective: To investigate the expression of hypoxia inducible factor-1α (HIF-1α) and cyclooxygenase-2 (COX-2) protein and mRNA in laryngeal squamous cell carcinoma in order to help the occurrence of laryngeal squamous cell carcinoma, Development and transfer mechanisms further clarified. Methods: The expressions of HIF-1α, COX-2 protein and mRNA in 76 cases of laryngeal squamous cell carcinoma and 9 cases of adjacent normal tissues were detected by immunohistochemistry and RT-PCR. The expressions of COX-2 and HIF-1α were detected by T stage and pathological grade HIF-1α, COX-2 protein and mRNA expression and clinicopathological parameters. Results: ①The expression of HIF-1α and COX-2 in laryngeal carcinoma specimens was significantly higher than that in adjacent normal tissues (P <0.05), and increased with the increase of T stage. The expression of HIF- 1α expression was significantly higher than well-differentiated laryngeal carcinoma; COX-2 protein expression and pathological grade has nothing to do. ② The positive expression rates of HIF-1α and COX-2 mRNA in laryngeal carcinoma tissues were 67.11% and 75.00%, respectively, which were significantly higher than those in adjacent normal tissues (P <0.05). The expression of COX-2 mRNA in adjacent normal tissues was not detected. The positive expression rates of HIF-1α and COX-2 mRNA in laryngeal squamous cell carcinoma were correlated with T stage (P <0.05). The expression of HIF-1α and COX-2 mRNA in laryngeal squamous cell carcinoma was increased with the increase of T stage, but not with the pathological grade (P> 0.05). ③ The positive rate of COX-2 in positive cases of HIF-1α protein expression was 78.72%. Conclusion: The expression of HIF-1α and COX-2 in laryngeal squamous cell carcinoma are increased and closely related, which may be one of the reasons for the development of laryngeal carcinoma.
