目的通过观察脑缺血后白脉软膏对大鼠脑组织p38MAPK及GAP-43蛋白表达的影响,探讨白脉软膏对脑组织的神经保护作用。方法60只雄性Wistar大鼠随机分为正常组、缺血组、白脉干预组,大脑中动脉线栓法制作局灶性脑缺血大鼠模型,分别于1、3、7、14d处死动物,采用免疫组织化学法观察各组脑组织p38MAPK和GAP-43蛋白的表达。结果p38MAPK缺血组的表达在7、14d分别为178.2±18.7、164.8±21.5,显著高于白脉组的153.2±21.9、120.2±18.6(P<0.01);GAP-43白脉组的表达在7、14d分别为62.2±6.4、55.0±7.6,也显著高于缺血组的35.7±4.0、33.0±3.7(P<0.01)。结论白脉软膏可调节脑缺血后p38MAPK和GAP-43的表达,并与损伤时间的变化有一定相关性,提示白脉软膏可能与损伤后神经元的再生、修复有关,有进一步研究的价值。
Objective To observe the effect of Baimao Ointment on the expression of p38MAPK and GAP-43 protein in rat brain after cerebral ischemia, and to explore the neuroprotective effect of Baimai ointment on brain tissue. Methods Sixty male Wistar rats were randomly divided into normal group, ischemic group, and white vein intervention group. The middle cerebral artery occlusion method was used to make focal cerebral ischemia rats. The animals were sacrificed at 1, 3, 7 and 14 days, respectively. Immunohistochemistry was used to observe the expression of p38MAPK and GAP-43 protein in brain tissue of each group. Results The expression of p38MAPK in ischemic group was 178.2±18.7 and 164.8±21.5 on 7th and 14th day respectively, which was significantly higher than that of 153.2±21.9 and 120.2±18.6 in white blood group (P<0.01). The expression of GAP-43 white vein group was On the 7th and 14th days, they were 62.2±6.4 and 55.0±7.6, respectively, which were also significantly higher than those of the ischemic group (35.7±4.0, 33.0±3.7, P<0.01). Conclusion Baiyin ointment can regulate the expression of p38MAPK and GAP-43 after cerebral ischemia, and has a certain correlation with the time of injury, suggesting that Baimao ointment may be related to the regeneration and repair of injured neurons. It has further research value. .
