目的:观察在心导管治疗时的辐射对内皮素合成及释放的影响及其作用机制。方法:1Gy60Coγ射线照射KrebsHenstlei(KH)液孵育的猴血管组织,并在KH液中加入不同药物干预,放射免疫法测定组织及KH液中内皮素含量。结果:1Gy60Coγ射线可刺激血管内皮素的合成与释放,维生素E、无钙孵育液、蛋白激酶C抑制剂polymyxinB和H7可有效抑制这种刺激作用,而维拉帕米和丝裂素活化蛋白激酶激酶阻滞剂PD098059对此种作用无影响。结论:辐射所致自由基的产生,细胞内[Ca2+]增加,蛋白激酶C系统的激活促进了ET的生成,在心导管介入治疗后并发症发生过程中,电离辐射可能起着一定作用
Objective: To observe the effect of radiation on the synthesis and release of endothelin in cardiac catheterization and its mechanism. Methods: The monkey blood vessels incubated with 1Gy60Coγ-ray in KrebsHenstlei (KH) solution were treated with different drugs. The content of endothelin in tissues and KH fluid was determined by radioimmunoassay. Results: 1Gy60Coγ-ray can stimulate the synthesis and release of endothelin, vitamin E, calcium-free incubation solution, protein kinase C inhibitors polymyxinB and H7 can effectively inhibit this stimulation, and verapamil and mitogen-activated protein kinase The kinase inhibitor PD098059 has no effect on this effect. CONCLUSIONS: The generation of free radicals induced by radiation, intracellular [Ca2 +] increase and activation of protein kinase C system promote the production of ET. Ionizing radiation may play a role in the complication after cardiac catheterization
