为了探讨以甘草查尔酮A衍生物(4’-甲基-2,4-羟基查尔酮)对Lewis肺癌细胞的增殖抑制作用和对细胞周期的影响,试验利用MTT、流式细胞术及蛋白免疫印迹法检测4’-甲基-2,4-羟基查尔酮对Lewis肺癌细胞增殖的抑制作用及机制。结果表明:与空白对照组比较,各剂量组(10,20,40μmol/L)的4’-甲基-2,4-羟基查尔酮能有效抑制Lewis肺癌细胞增殖(P<0.05),并可显著诱导其细胞凋亡(P<0.05),其作用呈时间浓度依赖性;4’-甲基-2,4-羟基查尔酮可使Lewis肺癌细胞的Bax蛋白和Caspase-3蛋白表达增高,而使Bcl-2蛋白表达降低。说明4’-甲基-2,4-羟基查尔酮能有效抑制Lewis肺癌细胞的增殖并诱导其细胞凋亡,其机制可能与其改变细胞凋亡的线粒体途径中的Bax、Bcl-2及Caspase-3蛋白的表达水平有关。
In order to investigate the inhibitory effect of licochalcone A derivative (4’-methyl-2,4-hydroxy-chalcone) on the proliferation of Lewis lung cancer cells and its effect on cell cycle, MTT, flow cytometry and Inhibitory effect of 4’-methyl-2,4-hydroxy-chalcone on the proliferation of Lewis lung carcinoma cells and its mechanism by Western blotting. The results showed that compared with the control group, 4’-methyl-2,4-hydroxychalcone at various doses (10, 20 and 40 μmol / L) could effectively inhibit the proliferation of Lewis lung cancer cells (P <0.05), and its effect was time-dependent. 4’-methyl-2,4-hydroxychalcone increased the expression of Bax and Caspase-3 in Lewis lung cancer cells , Leaving the Bcl-2 protein expression decreased. These results suggest that 4’-methyl-2,4-hydroxychetone can effectively inhibit the proliferation and induce the apoptosis of Lewis lung cancer cells, which may be related to the change of Bax, Bcl-2 and Caspase -3 protein expression level.
