本研究旨在高胰岛素浓度状态,比较两种不同结构的ATP敏感钾通道开放剂尼可地尔(Nic)及RP49356(RP)对离体兔胸主动脉环的舒血管作用。结果显示周Nic、RP舒张苯肾上腺素诱导的血管收缩无内皮依赖性,格列苯脲能完全阻断RP的作用,但仅部分抑制Nic的作用。亚申蓝仅对Nic的作用有部分抑制作用。Ins可加强RP的舒血管作用而对Nic的影响不明显。结果提示,RP、Nic在高胰岛素浓度状态下的血管舒张机制存在差异。
The purpose of this study was to investigate the vasodilatation of isolated rabbit thoracic aortic rings with Nicotine (Nic) and RP49356 (RP), an ATP-sensitive potassium channel opener mediated by two different structures. The results showed that there was no endothelium-dependent vasoconstriction induced by diastolic phenylephrine in Nic, RP and glibenclamide completely blocked the effect of RP, but only partially inhibited the effect of Nic. Asias Blue only partially inhibits Nic’s effect. Ins can enhance the vasodilating effect of RP while the effect of Nic is not obvious. The results suggest that there are differences in the mechanism of vasodilatation of RP and Nic in the condition of high insulin concentration.
