目的 研究cyclin D1基因在原发性肝细胞癌中的表达,并探讨其与P16及CDK4蛋白表达的关系。 方法 用原位杂交检测cyclin D1基因mRNA表达,免疫组化检测Cyclin D1蛋白表达。 结果 6例(14.3%)HCC可检测到cyclin D1基因过表达,5例属Ⅲ—Ⅳ级,1例属Ⅱ级。6例HCC均有CDK4蛋白表达,其中3例为P16蛋白阳性。 结论 ①cyclin D1表达增加只在小部分恶性程度高的HCC中起作用,可能与肿瘤侵袭有关。②在HCC中,cyclin D1基因过表达和(或)P16蛋白丢失均起重要作用。
Objective To investigate the expression of cyclin D1 gene in primary hepatocellular carcinoma and its relationship with P16 and CDK4 protein expression. Methods The expression of cyclin D1 mRNA was detected by in situ hybridization and the expression of Cyclin D1 protein was detected by immunohistochemistry. Results Overexpression of cyclin D1 gene was detected in 6 cases (14.3%) of HCC, 5 cases belonged to grade III-IV, and 1 case belonged to grade II. All 6 cases of HCC had CDK4 protein expression, of which 3 cases were positive for P16 protein. Conclusion 1 The increased expression of cyclin D1 only plays a role in a small proportion of highly malignant HCC, which may be related to tumor invasion. 2 In HCC, cyclin D1 gene overexpression and (or) P16 protein loss all play an important role.
