目的:以中药外用传统剂型“跌创散”为对照,评价“跌创散”的现代巴布制剂“跌创膏”促进骨缺损愈合药效作用,探讨新剂型跌创膏对骨折模型骨折部BMP-2、VEGF表达的影响,明确其促进骨折的愈合机制。方法:取兔龄3个月的雄性新西兰大耳白兔45只,造成桡骨骨折模型,随机分成模型组、跌创膏组、跌创散组。模型组自然愈合,跌创膏组外敷用最新中药提纯技术提纯并加透皮剂制成的跌创膏,跌创散组外敷传统剂型跌创散。于造模后1、2、4 w末行X线摄片,并采用免疫组化法检测骨折部骨痂BMP-2及VEGF表达。结果:各期跌创膏组和跌创散组X线评分结果与模型组比较有统计学意义,且跌创膏组与跌创散组比较有统计学意义;第2周跌创膏组与跌创散组的BMP-2、VEGF表达差异有统计学意义(P<0.05)。结论:跌创膏在骨折愈合过程中能促进骨痂BMP-2、VEGF表达,加快骨折愈合,其疗效优于传统剂型跌创散。
OBJECTIVE: To evaluate the efficacy of “traditional Chinese medicine preparation” or “Chuangsan Powder” as a contrast to evaluate the effect of modern Babu preparations or “Chuangao Paste” to promote the healing of bone defects, The effect of ointment on the expression of BMP-2 and VEGF in the fractured fracture model and the mechanism of its promotion of fracture healing. Methods: Forty-five male New Zealand white rabbits, 3 months old, were randomly divided into model group, Dacao cream group and Shichuang powder group. Model group of natural healing, or a set of topical cream group with the latest Chinese medicine purification technology and added transdermal agent made of or a wound cream, or Chuangsan group tops traditional Chinese medicine or Chuangsan scattered. X-ray radiography was performed at the end of 1, 2, 4 w after modeling, and immunohistochemistry was used to detect the expression of BMP-2 and VEGF in callus of fracture. Results: The results of X-ray score of each group of Shichuang ointment and that of Shouchuangsan were statistically significant compared with the model group, There were significant differences in BMP-2 and VEGF expression in the ascending group (P <0.05). CONCLUSION: ORG can promote the expression of BMP-2 and VEGF in callus during fracture healing and accelerate the healing of fractures. The effect is better than that of traditional one.
