目的 比较 CD71单染和 CD71、Hoechst332 5 8(HO2 5 8)双染对单个核细胞的标记情况 ,并用于对孕妇外周血中有核红细胞 (nucleated red blood cells,NRBCs)的分选。方法 选用红细胞特异性抗体CD71、核染料 HO2 5 8对正常孕妇外周血、轻 (或 )中度妊娠高血压综合征 (简称妊高症 )患者外周血、初生婴儿脐血单个核细胞进行标记 ,并结合流式细胞术对有核红细胞进行分选。结果 用 CD71和 HO2 5 8进行双染色标记 ,正常孕妇、妊高症患者、脐血单个核细胞的阳性标记率明显低于 CD71单染色标记 (t检验 ,P<0 .0 1) ,并成功地用于流式细胞术分选出孕妇外周血中的 NRBCs。结论 采用 CD71和 HO2 5 8双染色标记 ,可以排除成熟红细胞、血小板、网织红细胞 ,提高 NRBCs的特异性标记和分选纯度
OBJECTIVE: To compare the labeling of mononuclear cells by CD71 single staining and CD71, Hoechst332 58 (HO2 58) double staining and to use in the sorting of nucleated red blood cells (NRBCs) in peripheral blood of pregnant women. Methods Peripheral blood and neonatal cord blood mononuclear cells of normal pregnant women with mild or moderate pregnancy-induced hypertension (PIH) were labeled with erythrocyte-specific antibody CD71 and nuclear dye HO2 58, Combined with flow cytometry of nucleated red blood cells were sorted. Results The double staining of CD71 and HO2 58 showed that the positive labeling rates of normal pregnant women, patients with pregnancy-induced hypertension and umbilical cord blood mononuclear cells were significantly lower than those of CD71 single-stained staining (t test, P <0.01) For flow cytometry sorting of peripheral blood of pregnant women NRBCs. Conclusion CD71 and HO2 58 double staining can eliminate mature erythrocytes, platelets, reticulocytes and improve the specific labeling and sorting purity of NRBCs
