目的通过观察穴位埋线对大鼠胶元诱导型关节炎(CIA)模型血清Treg/Th17相关细胞因子及滑膜组织病理形态学的影响,从细胞因子角度以及组织病理形态学改变探讨其的作用机制。方法建立Ⅱ型胶原诱导大鼠胶原性关节炎模型,造模成功的大鼠随机分为模型组、来氟米特组、穴位埋线治疗组、来氟米特+穴位埋线治疗组。动态观察大鼠一般情况、各项关节炎活动指标、HE染色观察滑膜病理改变、ELISA法检测血清中IL-6、IL-17和TGF-β1含量。结果与模型组比较,各治疗组均能有效改善CIA大鼠关节肿胀以及关节炎指数;提高血清中TGF-β1含量和降低血清中IL-6、IL-17含量,各治疗组能有效减轻CIA模型滑膜组织关节炎炎症及病理形态的改变。结论穴位埋线治疗具有免疫调节作用,其可能调节Th17/Treg细胞平衡而显著减轻关节炎症和破坏。
Objective To investigate the effects of catgut embedding on serum Treg / Th17-related cytokines and synovial histopathology in rats with collagen-induced arthritis (CIA), and to explore its role in cytokines and histopathological changes mechanism. Methods Collagen induced arthritis model induced by type Ⅱ collagen was established in rats. The successful rats were randomly divided into model group, leflunomide group, catgut embedding therapy group and leflunomide + catgut implantation group. Dynamic observation of the general situation of rats, the various indicators of arthritis, HE staining observed synovial pathological changes, serum IL-6, IL-17 and TGF-β1 levels were measured by ELISA. Results Compared with the model group, each treatment group can effectively improve the joint swelling and arthritis index in CIA rats; increase the content of TGF-β1 in serum and decrease the content of IL-6 and IL-17 in serum in CIA rats; Changes of inflammation and pathological changes in synovial tissue of arthritis model. Conclusion Catgut embedding therapy has the immunomodulatory effect, which may regulate Th17 / Treg cell balance and significantly reduce joint inflammation and destruction.
