【目的】观察冰片对兔角膜上皮细胞的损伤作用。【方法】以浓度分别为100、200、400μg/m L的冰片作用于兔角膜上皮细胞,采用四甲基偶氮唑盐(MTT)法检测细胞活性,Annexin V-异硫氰酸荧光素/碘化丙啶(Annexin V-FITC/PI)双染法流式细胞仪检测细胞凋亡率;Real-time PCR法检测细胞凋亡相关基因Caspase-3 m RNA的表达。【结果】不同浓度的冰片均可抑制兔角膜上皮细胞的活性;与正常对照组比较,冰片组可增加兔角膜上皮细胞的凋亡率,增强Caspase-3 m RNA的表达(P<0.05或P<0.01)。【结论】冰片能抑制兔角膜上皮细胞的生长,诱导细胞发生凋亡,其机制与增强凋亡相关基因Caspase-3m RNA的表达有关。
【Objective】 To observe the damage of borneol on rabbit corneal epithelial cells. 【Methods】 The rabbit corneal epithelial cells were treated with 100, 200 and 400 μg / mL of borneol, respectively. The cell viability was measured by MTT assay. Annexin V-fluorescein isothiocyanate / Apoptosis rate was detected by flow cytometry with Annexin V-FITC / PI double staining method. Caspase-3 mRNA expression was detected by Real-time PCR. 【Results】 The results showed that the different concentrations of borneol could inhibit the activity of rabbit corneal epithelial cells. Compared with the normal control group, borneol increased the apoptotic rate of rabbit corneal epithelial cells and increased the expression of Caspase-3 mRNA (P <0.05 or P <0.01). 【Conclusion】 Borneol can inhibit the growth of rabbit corneal epithelial cells and induce cell apoptosis. The mechanism is related to the enhancement of the expression of Caspase-3m RNA.
