[目的]探讨姬松茸多糖对哮喘模型小鼠气道炎症和Th2类细胞因子水平的影响.[方法]取雌性Balb/c小鼠40只,随机分为正常对照组、哮喘模型组和姬松茸多糖小、大剂量组,姬松茸多糖小、大剂量组分别灌胃给予姬松茸多糖200,400mg/kg.采用酶联免疫吸附法和蛋白质印迹法检测支气管肺泡灌洗液(BALF)中白细胞介素(IL)-4,IL-5,IL-13含量,并观察BALF中炎症细胞计数和肺组织病理学改变.[结果]与正常对照组比较,哮喘模型组小鼠BALF中炎症细胞计数及IL-4,IL-5,IL-13水平均明显升高(P<0.05).与哮喘模型组比较,姬松茸多糖小、大剂量组BALF中炎症细胞计数及IL-4,IL-5,IL-13水平均明显降低(P<0.05).姬松茸多糖可明显减轻哮喘模型小鼠肺组织病理学改变.[结论]姬松茸多糖具有抗哮喘作用,其机制可能与下调Th2炎症细胞因子、减少炎症细胞浸润有关.
[Objective] To explore the effect of Agaricus Blazei polysaccharide on airway inflammation and Th2 cytokine in asthmatic mice. [Methods] Forty female Balb / c mice were randomly divided into normal control group, asthmatic model group and Agaricus blazei The polysaccharides small and large dose group, Agaricus blazei polysaccharides small and high dose group were given intragastric administration of Agaricus blazei polysaccharide 200,400mg / kg. ELISA and Western blot were used to detect interleukin in bronchoalveolar lavage fluid (BALF) (IL) -4, IL-5 and IL-13 in BALF and the changes of inflammatory cells and pathological changes in BALF were observed. [Results] Compared with the normal control group, the number of inflammatory cells and IL (P <0.05) .Compared with the asthma model group, the number of inflammatory cells and the levels of IL-4, IL-5, IL (P <0.05) .Conclusion: Polysaccharides of Agaricus Blazei Murrill has the anti-asthmatic effect, which may be related to the down-regulation of Th2 inflammatory cytokines and the decrease of Inflammatory cell infiltration.
