目的探讨高浓度葡萄糖对体外培养的恒河猴脉络膜-视网膜内皮细胞(RF/6A)的损伤及姜黄素对此的保护作用。方法 RF/6A为3组培养(n=6):对照组(DMEM完全培养基+10%胎牛血清),高浓度葡萄糖组(DMEM完全培养基+10%胎牛血清+30mmol/L葡萄糖),姜黄素-葡萄糖组(DMEM培养基+10%胎牛血清+30mmol/L葡萄糖+30μmol/L姜黄素)。以噻唑蓝(MTT)法检测分组培养1、3、5d后各组细胞活力的变化,检测培养5d后各组细胞丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性。结果高浓度葡萄糖组RF/6A活力逐渐降低,呈现明显的时间依赖性,与对照组相比高浓度葡萄糖组RF/6A活性降低明显(P<0.01)。姜黄素-葡萄糖组RF/6A活力无明显变化,与对照组无统计学差异(P>0.05)。氧化应激水平检测显示,与对照组相比,分组培养5d后,高浓度葡萄糖组MDA含量增加(P<0.01),SOD活力降低(P<0.01)。姜黄素-葡萄糖组MDA含量及SOD活力均无明显变化(P>0.05)。结论姜黄素可抑制RF/6A氧化应激水平,防止高浓度葡萄糖对细胞造成损伤。
Objective To investigate the injury of rhesus choroid-retina endothelial cells (RF / 6A) cultured in vitro by high concentration glucose and the protective effect of curcumin. Methods RF / 6A was cultured in 3 groups (n = 6): control group (DMEM complete medium + 10% fetal bovine serum), high concentration glucose group (DMEM complete medium + 10% fetal bovine serum + 30 mmol / , Curcumin-glucose group (DMEM medium + 10% fetal bovine serum + 30 mmol / L glucose + 30 μmol / L curcumin). The cell viability in each group was detected by MTT assay after 1, 3 and 5 days. The content of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD) in each group were detected 5 days after culture. Results Compared with the control group, the RF / 6A activity of high glucose group decreased significantly (P <0.01). Curcumin - glucose group RF / 6A activity no significant change, and the control group no significant difference (P> 0.05). Oxidative stress test showed that compared with the control group, MDA content increased (P <0.01) and SOD activity decreased (P <0.01) 5d after cultured in high concentration glucose group. Curcumin - glucose group MDA content and SOD activity had no significant change (P> 0.05). Conclusion Curcumin can inhibit the level of oxidative stress induced by RF / 6A and prevent the injury caused by high concentration of glucose.
