目的探讨重组人生长激素(r-hGH)对人乳腺癌细胞株MCF-7(MCF-7)有无促进增殖和分化作用,以及r-hGH与5-氟尿嘧啶(5-FU)联合应用对MCF-7的化疗协调作用。方法以不同浓度r-hGH,以及不同浓度的r-hGH联合不同浓度的5-FU分别加入处于对数生长期的MCF-7中进行体外培养,48h后流式细胞仪检测细胞周期的变化,与对照组进行比较。结果①不同浓度的r-hGH与MCF-7作用后,其细胞周期的G0~G1期、S期、G2~M期的细胞百分比变化,差异均无统计学意义(P>0.05);②不同浓度的5-FU与MCF-7作用后,其细胞周期的G0~G1期细胞百分比显著增高(P<0.05),S期显著下降(P<0·01),G2~M期差异无统计学意义(P>0.05);③不同浓度的r-hGH联合不同浓度的5-FU与MCF-7作用后,其细胞周期的G0~G1期细胞百分比显著增高(P<0.01),S期和G2~M显著降低(P<0.01,P<0.05)。结论单独应用r-hGH对MCF-7的细胞周期无明显影响;r-hGH与5-FU联合应用对MCF-7的化疗具有协调作用,r-hGH能增强5-FU对MCF-7的化疗效果。
Objective To investigate the effect of recombinant human growth hormone (r-hGH) on the proliferation and differentiation of human breast cancer cell line MCF-7 (MCF-7) and the combination of r-hGH and 5-fluorouracil -7 chemotherapy coordination effect. Methods Different concentrations of r-hGH and different concentrations of r-hGH combined with different concentrations of 5-FU were added to the logarithmic growth phase of MCF-7 cultured in vitro, 48h after the change of cell cycle detected by flow cytometry, Compared with the control group. Results ① The percentages of cells in G0 ~ G1 phase, S phase and G2 ~ M phase were not significantly different between different concentrations of r-hGH and MCF-7 (P> 0.05) (P <0.05), S phase decreased significantly (P <0.01), and there was no significant difference in G2 ~ M phase between 5-FU and MCF-7 (P> 0.05); ③The percentage of cells in G0 ~ G1 phase increased significantly (P <0.01) after different concentrations of r-hGH combined with different concentration of 5-FU and MCF-7, ~ M was significantly lower (P <0.01, P <0.05). Conclusion r-hGH alone has no effect on the cell cycle of MCF-7. The combination of r-hGH and 5-FU has a synergistic effect on MCF-7 chemotherapy. R-hGH can enhance the chemotherapeutic effect of 5-FU on MCF-7 effect.
