方法:针对大肠癌围手术期患者的主要中医证候阴虚证,采集符合条件病例的大肠癌组织及癌周正常组织,采用罗氏公司含12×135K个基因的人全基因组芯片检测大肠癌组织差异表达基因,筛选并分析阴虚证癌组织组和癌周正常组织组的差异表达基因,利用Pathway对相关基因进行生物信息学分析。结果:得出大肠癌围手术期患者阴虚证癌组织组和癌周正常组织组表达的差异基因共4869个,包括上调基因1574个,下调基因3295个。经检索数据库Gene Bank,获取基因名称等相关信息。进行Pathway分析,筛选出阴虚证癌组织组和癌周正常组织组之间具有显著差异表达基因主要涉及的信号转导通路。结论:大肠癌围手术期阴虚证癌组织组与癌周正常组织组比较,存在组织差异基因表达特征图谱,主要涉及主要涉及炎症反应,细胞内吞功能,矿物质吸收,酮体合成与分解功能,免疫功能等相关基因。其主要信号转导通路包括MAPK信号通路及p53信号通路等。
Methods: According to the yin deficiency syndrome of major TCM syndromes of perioperative patients with colorectal cancer, colorectal cancer tissues and normal tissues around the cancer were collected. Roche’s human genome-wide cDNA microarray containing 12 × 135K genes was used to detect colorectal cancer Differentially expressed genes were screened and analyzed for differentially expressed genes in yin deficiency cancer tissues and peritumoral normal tissues. Bioinformatic analysis of related genes was performed using Pathway. RESULTS: A total of 4869 differentially expressed genes were found in patients with colorectal cancer during the operation of yin deficiency syndrome and normal tissue, including 1574 up-regulated genes and 3295 down-regulated genes. Retrieve the gene name and other related information by searching Gene Bank. Pathway analysis was performed to screen out the signal transduction pathways that were mainly involved in differentially expressed genes between the Yin-deficiency and cancer-normal tissues. CONCLUSIONS: There are tissue differential gene expression profiles in perioperative Yin-deficiency syndromes of colorectal cancer, which mainly relate to inflammatory response, endocytosis, mineral absorption, ketone body synthesis and decomposition Function, immune function and other related genes. Its major signal transduction pathways include MAPK signaling pathway and p53 signaling pathway.
