目的 观察抗炎因子白细胞介素 10 (IL 10 )在血管成形术后的表达以探讨IL 10在血管损伤反应过程中的意义。方法 采用RT PCR、WesternBlot及免疫组化技术观察了大鼠主动脉球囊损伤后IL 10mRNA水平、蛋白表达及其蛋白分布的动态变化。结果 正常大鼠主动脉无IL 10的表达 ,在球囊损伤后第 1、7和 14天时 ,IL 10均出现转录 ,蛋白的表达量上调 ,生成的IL 10蛋白主要分布在血管内膜及中膜的血管平滑肌细胞 (VSMC)的胞浆和细胞外基质中。结论 大鼠主动脉球囊损伤后IL 10出现高表达 ,提示IL 10可能参与了血管损伤后局部炎症反应的修复过程
Objective To observe the expression of interleukin 10 (IL 10) after angioplasty in order to investigate the significance of IL 10 in the process of vascular injury. Methods RT-PCR, Western Blot and immunohistochemistry were used to observe the changes of IL-10 mRNA, protein expression and protein distribution after aortic balloon injury in rats. Results No IL 10 was expressed in the aorta of normal rats. At days 1, 7 and 14 after balloon injury, IL 10 was transcribed and the expression of protein was up-regulated. The produced IL 10 protein mainly distributed in the intima and intima Membranes of vascular smooth muscle cells (VSMCs) in the cytoplasm and extracellular matrix. Conclusions High expression of IL-10 occurs after aortic balloon injury in rats, suggesting that IL-10 may be involved in the repair process of local inflammatory response after vascular injury
