目的探讨重复、长时间正加速度(+Gz)暴露后正常和心肌缺血兔缝隙连接蛋白43(connexin 43,Cx43)表达的变化。方法新西兰大白兔60只,随机分为对照组、加速度组、缺血组、缺血并加速度组,每组15只。采用+Gz暴露法,暴露条件:起始+8 Gz,G值增长率约为0.5 G/s,峰值持续时间为1 min,间隔为3 min,一共3次,1次/d,暴露时间为1周。造模结束后处死动物留取左心室前壁心肌组织标本做Western blot分析,检测Cx43表达的变化。结果 Western blot结果显示,与对照组比较,心室肌细胞间Cx43水平降低的程度由低至高的顺序依次为:加速度组、缺血组、加速度并缺血组。结论心肌细胞Cx43的降低可能参与加速度诱发缺血心肌产生心律失常的机制。
Objective To investigate the changes of connexin 43 (Cx43) expression in normal and myocardial ischemic rabbits after repeated, long-term positive acceleration (+ Gz) exposure. Methods Sixty New Zealand white rabbits were randomly divided into control group, acceleration group, ischemia group, ischemia and acceleration group, 15 rats in each group. With the + Gz exposure method, the exposure conditions were as follows: the initial +8 Gz, the G value increasing rate was about 0.5 G / s, the peak duration was 1 min, the interval was 3 min, a total of 3 times, 1 time / d, 1 week. After modeling, animals were sacrificed and the left anterior myocardium was taken for Western blot analysis to detect the change of Cx43 expression. Results Western blot results showed that compared with the control group, the decreasing order of Cx43 levels in ventricular myocytes was acceleration group, ischemia group, acceleration and ischemia group. Conclusions The decrease of Cx43 in cardiomyocytes may be involved in the mechanism of acceleration-induced arrhythmia in ischemic myocardium.
