目的:研究五指莲重楼根茎中的化学成分。方法:采用溶剂法提取,利用正相硅胶色谱、葡聚糖凝胶SephadexLH-20、反相制备色谱等手段进行分离纯化,并通过现代波谱技术进行结构鉴定。结果:从乙酸乙酯层和正丁醇层中分离得到14个化合物,分别鉴定为胡萝卜苷(1),豆甾醇-3-O-β-D-吡喃葡萄糖苷(2),β-蜕皮激素(3),偏诺皂苷元-3-O-α-L-呋喃阿拉伯糖基(1→4)-[α-L-吡喃鼠李糖基(1→2)]-β-D-吡喃葡萄糖苷(4),薯蓣皂苷元-3-O-α-L-吡喃鼠李糖基(1→4)-α-L-吡喃鼠李糖基(1→4)-[α-L-吡喃鼠李糖基(1→2)]-β-D-吡喃葡萄糖苷(5),山柰酚(6),芦丁(7),杨梅素(8),4,2′,4′-trihydroxychalcone(9),异鼠李素-3-O-β-D-吡喃葡萄糖苷(10),异鼠李素-3-O-芸香糖苷(11),异鼠李素-3-O-龙胆双糖苷(12),5,5′,7-三羟基-4′,4,7″-三甲氧基-[3′-8″]双黄酮(13),5,5′,7,4′,4,7″-六羟基-[3′-8″]双黄酮(14)。结论:化合物1~3,6~14为首次从该种植物中分离得到,其中化合物7~9,13和14为首次从该属植物中分离得到。
Objective: To study the chemical composition of Rhizoma Rhei Methods: Solvent extraction and purification by normal phase silica gel, Sephadex LH-20 and reversed-phase preparative chromatography were used to identify the structures of the compounds. Results: Fourteen compounds were isolated from the ethyl acetate and n-butanol layers and identified as daucosterol (1), stigmasterol-3-O-β-D-glucopyranoside (2) (1 → 4) - [α-L-rhamnopyranosyl (1 → 2)] - β-D-D-glucopyranoside (3) (4), diosgenin-3-O-α-L-rhamnopyranosyl (1 → 4) -α-L-rhamnopyranosyl (1 → 2)] - β-D-glucopyranoside (5), kaempferol (6), rutin (7), myricetin (8), 4,2 ’ , 4’-trihydroxychalcone (9), isorhamnetin-3-O-β-D-glucopyranoside (10), isorhamnetin-3-O- rutinoside (11) (12), 5,5 ’, 7-trihydroxy-4’, 4,7 “-trimethoxy- [3’-8”] biflavone (13), 5,5 ’, 7,4’, 4,7 “-hexahydroxy- [3’-8”] biflavone (14). Conclusion: Compounds 1 ~ 3 and 6 ~ 14 were isolated from this plant for the first time. Among them, compounds 7 ~ 9, 13 and 14 were isolated from this genus for the first time.
