为研究葡萄糖、精氨酸对新生大鼠胰β细胞中分泌颗粒(β细胞颗粒)的影响,选用Wistar新生大鼠,自出生后第2d起腹腔注射葡萄糖或葡萄糖加精氨酸,共6d。分别于出生后第10d、20d剖腹取胰腺,进行透射电镜观察,对β细胞颗粒进行立体定量分析,并根据体视学有关原理,从β细胞颗粒的不同大小切面的频数分布,重建其球体大小的频数分布。结果显示,葡萄糖组的β细胞颗粒仅有少量增加,其他参数较对照组无明显差异。葡萄糖加精氨酸组的体密度、数面密度、数体密度、平均直径、平均体积、平均表面积均比对照组显著增大。这提示:用葡萄糖加精氨酸可促进新生大鼠胰β细胞合成、贮存与分泌胰岛素。
In order to study the effects of glucose and arginine on secretory granules (β-cell granules) in rat pancreatic β cells, Wistar neonatal rats were used to inoculate glucose or glucose and arginine intraperitoneally on the 2nd day after birth for 6 days. The pancreas were harvested on the 10th day and the 20th day after birth respectively. The pancreas was dissected and observed by transmission electron microscopy. The β-cell pellets were analyzed by stereotactic method. According to the principle of stereology, the size distribution of the β-cell pellets Frequency distribution. The results showed that only a small amount of β-cell particles in glucose group increased, other parameters no significant difference compared with the control group. Glucose plus arginine group body density, number area density, number density, average diameter, average volume, average surface area than the control group increased significantly. This suggests that glucose and arginine can promote pancreatic β cell synthesis, storage and secretion of insulin in neonatal rats.
