Micro RNAs(miRNAs)是内源性的调节基因转录后水平的一类小分子RNA。研究表明miRNA164/NAC在模式植物拟南芥中参与调控植物的生长发育以及对生物和非生物胁迫的耐受性,但目前在马铃薯中关于miRNA164/NAC的表达模式的研究尚未见报道。本研究利用马铃薯中已知的miRNA164序列,通过在线工具ps RNATarget对其靶基因进行预测得到3个靶基因。通过生物信息学分析发现,马铃薯miRNA164的3个靶基因均属于NAC转录因子家族的NAM亚家族,其调控模式与拟南芥中的类似,具有很高的保守性。为了进一步实验验证,利用人工miRNA(artificial miRNA,amiRNA)技术构建了马铃薯amiRNA164表达载体,有助于后期通过遗传转化方法研究miRNA164的调控机理。
Micro RNAs (miRNAs) are a class of small RNAs that endogenously regulate the post-transcriptional level of a gene. Studies have shown that miRNA164 / NAC is involved in the regulation of plant growth and development and tolerance to biotic and abiotic stresses in the model plant Arabidopsis thaliana. However, the current research on the expression pattern of miRNA164 / NAC in potato has not been reported yet. In this study, three target genes were predicted by using the miRNA164 sequence known in potato and their target genes were predicted by on-line tool ps RNATarget. Bioinformatics analysis showed that all three target genes of potato miRNA164 belong to the NAM subfamily of NAC transcription factor family, and their regulation pattern is similar to that in Arabidopsis thaliana, which is highly conserved. In order to further validate the experiment, artificial amRNA (artificial miRNA, amiRNA) was used to construct the amiRNA164 expression vector, which is helpful to study the regulation mechanism of miRNA164 through genetic transformation later.
