目的:探讨威灵仙水提物对阿霉素诱导耐药的人甲状腺未分化癌细胞株HTh74Rdox增殖的影响及机制。方法:用不同浓度威灵仙水提物处理HTh74Rdox后观察细胞形态;噻唑蓝(MTT)比色法检测威灵仙水提物对HTh74Rdox细胞的增殖抑制作用;流式细胞术检测HTh74Rdox细胞凋亡;蛋白质免疫印迹法(Western blot)检测凋亡相关半胱氨酸蛋白酶-3(Caspase-3)及其活性片段(cleaved-Caspase-3),B淋巴细胞瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)的表达。结果:威灵仙水提物对HTh74Rdox细胞的增殖有明显抑制作用,联合阿霉素后可减少阿霉素剂量;威灵仙水提物可显著诱导细胞凋亡(P<0.01);且能明显提高cleaved-Caspase-3/Caspase-3,Bax/Bcl-2(P<0.05)。结论:威灵仙水提物可以抑制HTh74Rdox细胞的增殖,其机制可能与提高cleaved-Caspase-3/Caspase-3,Bax/Bcl-2有关;联合化疗药物使用具有减毒增效的作用。
OBJECTIVE: To investigate the effect and mechanism of aqueous extract of Clematis on the proliferation of doxorubicin-resistant human thyrotropin-releasing cell line HTh74Rdox. Methods: The HTh74Rdox cells were treated with different concentrations of Clematis water extract, the cell morphology was observed, the proliferation of HTh74Rdox cells was detected by MTT colorimetric assay, the apoptosis of HTh74Rdox cells was detected by flow cytometry, Western blot was used to detect the expression of Caspase-3, cleaved-Caspase-3, Bcl-2 and Bcl-2 Related X protein (Bax) expression. Results: Water extract of Clematis significantly inhibited the proliferation of HTh74Rdox cells. Combination of doxorubicin and doxorubicin reduced the dose of doxorubicin. Clematis water extract could significantly induce apoptosis (P <0.01), and significantly increased cleaved-Caspase-3 / Caspase-3, Bax / Bcl-2 (P <0.05). Conclusion: Clematis water extract can inhibit the proliferation of HTh74Rdox cells. The mechanism may be related to the increase of cleaved-Caspase-3 / Caspase-3 and Bax / Bcl-2.
