目的了解北京市丰台区污水中埃可病毒6型(echovirus 6,ECHO6)的基因特征。方法选择吴家村污水处理厂为监测点,2012年1~12月,每月采集监测点污水进行肠道病毒(enterovirus,EV)分离和鉴定,并对分离到的ECHO6进行VP1编码区核苷酸序列测定和分析。结果在吴家村污水处理厂监测点,共分离到64株病毒,其中ECHO6病毒12株,在非脊髓灰质炎(脊灰)肠道病毒(non-poliomyelitis entero-viruses,NPEV)分离株中比例最高。选择10株ECHO6进行VP1编码区核苷酸序列分析,结果表明10株病毒的VP1区核苷酸同源性为92.9%~100.0%,与D5基因型代表株的VP1区核苷酸同源性最高,为88.7%~94.2%。进化树分析结果显示,北京市丰台区ECHO6分离株与D5基因型代表株处于同一分支,并在D5进化分支的不同簇中。结论北京市丰台区污水中ECHO6分离株为D5基因型并在D5进化分支的不同簇中,并为丰台区人群肠道病毒的监测提供重要的补充信息。
Objective To understand the genetic characteristics of echovirus 6 (ECHO6) in Fengtai District of Beijing. Methods Wujiacun WWTP was selected as the monitoring point. From January to December in 2012, sewage was collected from monitoring points every month for the isolation and identification of enterovirus (EV). The isolated ECHO6 was subcloned into VP1 coding region Sequencing and analysis. Results A total of 64 viruses were isolated from the monitoring sites of Wujiacun WWTP. Among them, 12 strains of ECHO6 virus were the highest among non-polioomyelitis entero-viruses (NPEV) isolates . The nucleotide sequence of the VP1 coding region of 10 strains of ECHO6 was selected. The results showed that the VP1 nucleotide homology of the 10 strains of viruses was 92.9% -100.0%, which was homologous to the VP1 region of D5 genotype Highest, 88.7% ~ 94.2%. Phylogenetic tree analysis showed that the ECHO6 isolate in Fengtai District of Beijing was in the same branch as the representative D5 genotype and was found in different clusters of D5 evolutionary branch. Conclusion The ECHO6 isolates in Fengtai District of Beijing are D5 genotypes and belong to different clusters of D5 evolutionary branch, and provide important supplementary information for the monitoring of enterovirus in Fengtai district.
