目的:研究人脑胶质瘤组织中丝裂原激活蛋白激酶(MAPK)的表达及活化情况。方法:应用免疫组织化学技术观察新鲜冻存的16例脑胶质瘤组织标本(其中Ⅱ级9例,Ⅲ级7例)和5例脑外伤内减压术后正常脑组织中MAPK及磷酸化MAPK(p-MAPK)的表达情况。结果:MAPK阳性表达位于胞质;p-MAPK位于胞质和胞核,且胞核阳性信号较强。胶质瘤Ⅱ级、Ⅲ级和正常脑组织中MAPK阳性区域面积百分率分别为(37.78±4.05)%、(53.58±1.60)%和(23.12±2.43)%,p-MAPK阳性区域面积百分率分别为(36.91±3.85)%、(53.14±3.47)%和(18.61±3.30)%。胶质瘤组织中MAPK及p-MAPK阳性区域面积百分率均高于正常脑组织(P<0.05);胶质瘤Ⅲ级者高于Ⅱ级者(P<0.05)。结论:MAPK及其异常活化与人脑胶质瘤的发生发展有关。
Objective: To study the expression and activation of mitogen-activated protein kinase (MAPK) in human glioma tissue. Methods: Immunohistochemistry was used to detect the expression of MAPK and phosphorylation in 16 fresh glioma samples (9 in grade Ⅱ, 7 in grade Ⅲ) and 5 normal brain tissue after decompression in traumatic brain injury MAPK (p-MAPK) expression. Results: The positive expression of MAPK was localized in the cytoplasm. The positive expression of p-MAPK was located in the cytoplasm and nucleus. The percentages of MAPK positive area in grade Ⅱ, grade Ⅲ and normal brain tissue were (37.78 ± 4.05)%, (53.58 ± 1.60)% and (23.12 ± 2.43)% respectively, and the percentage of positive area of p-MAPK were (36.91 ± 3.85)%, (53.14 ± 3.47)% and (18.61 ± 3.30)%, respectively. The percentages of positive area of MAPK and p-MAPK in glioma tissues were higher than those in normal brain tissues (P <0.05). The gliomas in grade Ⅲ were higher than those in grade Ⅱ (P <0.05). Conclusion: MAPK and its abnormal activation are related to the occurrence and development of human glioma.
