利用随机扩增多态性DNA(Randomly amplified polymorphic DNA,RAPD)分子标记和集团分离分析法(Bulked segregation analysis,BSA)分析技术,对番木瓜(Carica papaya L.)F_2代分离群体和抗番木瓜环斑型花叶病毒(Papaya ringspot virus,PRSV)突变体LK-1的抗PRSV基因进行连锁分析,从250个随机引物中筛选出了与抗PRSV基因相连锁的3个RAPD标记,连锁分析表明,S1366、S7058和S7125与抗PRSV基因(rys)的遗传距离分别为1.9 cM、2.4 cM和12.2 cM,其中S1366和S7058两个标记在抗PRSV基因的一侧,S7125在另一侧。
The random amplified polymorphic DNA (RAPD) markers and bulked segregation analysis (BSA) were used to analyze the segregation population of F_2 progenies of Carica papaya L., The PRSV gene of the mutant LK-1 from the Papaya ringspot virus (PRSV) was analyzed by linkage analysis. Three RAPD markers linked to the anti-PRSV gene were screened out from 250 random primers. The linkage analysis showed that The genetic distances of S1366, S7058 and S7125 to the anti-PRSV gene (rys) were 1.9 cM, 2.4 cM and 12.2 cM respectively, of which two markers S1366 and S7058 were on the anti-PRSV gene side and the S7125 was on the other side.
