目的探讨低氧对新生鼠肺组织内皮素转化酶-1(ECE-1)表达的影响。方法日龄5~7天的Wistar新生鼠30只,随机分为对照组和缺氧组各15只。对照组置室温空气中,缺氧组给予低氧干预(氧浓度5%~6%)6h,开胸取肺,观察肺大体改变,切片行HE染色观察病理学改变,采用免疫组织化学染色检测肺组织中ECE-1的表达。结果对照组肺外观无明显改变,缺氧组呈现肺水肿改变;光镜下缺氧组可见肺泡间隔增厚,间质水肿,毛细血管轻度充血;免疫组织化学染色显示ECE-1在缺氧组表达增强(0.404±0.029),与对照组(0.310±0.031)比较差异有统计学意义(P<0.05)。结论低氧可使新生鼠肺组织ECE-1表达增强,ECE-1可通过促进内皮素-1加快转化参与低氧所致肺组织损伤的形成过程。
Objective To investigate the effect of hypoxia on the expression of endothelin-converting enzyme-1 (ECE-1) in the lung of newborn rats. Methods Thirty Wistar newborn rats aged 5 to 7 days were randomly divided into control group and hypoxia group. In the control group, the hypoxia group was given hypoxia intervention (oxygen concentration 5% -6%) for 6 hours, the lungs were opened by thoracotomy, the changes of the lungs were observed, and the pathological changes were observed by HE staining. Immunohistochemical staining ECE-1 expression in lung tissue. Results The lung appearance of the control group showed no significant change, and the changes of pulmonary edema appeared in the hypoxia group. Alveolar septum thickening, interstitial edema and capillary hyperemia were found in the hypoxia group under light microscope. Immunohistochemical staining showed that the expression of ECE- (0.404 ± 0.029), which was significantly different from that of the control group (0.310 ± 0.031) (P <0.05). Conclusion Hypoxia can enhance the expression of ECE-1 in lung tissue of neonatal rats. ECE-1 may be involved in the process of lung injury induced by hypoxia by accelerating the transformation of endothelin-1.
