从新生牛主动脉分离获得单个内皮细胞 ,培养于微孔滤膜上直至形成致密单层。通过灌流 hanks液或含5 g/ L 白蛋白的 hanks液后 ,测定经 10 - 4 m ol/ L 组胺溶液处理或经 10 - 4 mol/ L 组胺溶液加 10 - 4 g/ m L 当归补血汤处理的液体滤过系数 (Kf )、液体滤过流量 (Jv)和蛋白质渗透压反射系数 (σ)。结果表明内皮单层经组胺处理后 ,Kf 和 Jv降低 ,σ升高 ;而当归补血汤能够抑制因组胺造成的 Kf 和 Jv降低 ,以及 σ升高。说明组胺能够引起血管内皮单层通透性的增加 ,而当归补血汤能够减轻组胺造成的内皮单层通透性的增加 ;其作用机制需进一步研究。
Single endothelial cells were isolated from newborn bovine aorta and cultured on microporous filters until a dense monolayer was formed. After perfusion of the Hanks solution or Hanks’ solution containing 5 g/L of albumin, determine after treatment with 10 - 4 mol/L histamine solution or 10 - 4 mol/L histamine solution plus 10 - 4 g/m L. The blood filtration coefficient (Kf), liquid filtration flux (Jv) and protein osmotic pressure reflection coefficient (σ) of the blood soup treatment. The results showed that after the histamine treatment of endothelial monolayer, Kf and Jv decreased and σ increased; while Danggui Buxue Decoction could inhibit the decrease of Kf and Jv caused by histamine, and the increase of σ. It is suggested that histamine can increase the permeability of vascular endothelial monolayer, and Danggui Buxue Decoction can reduce the increase of histamine-induced endothelial monolayer permeability; its mechanism of action needs further study.
