目的观察珍宝丸预处理对不同时间点脑缺氧新生乳鼠大脑脑室的保护作用。方法取新生1天Wistar大鼠210只,雌雄不限,随机分对照组、模型组和珍宝丸组各70只,每组又分为7个时间点0.5、24、48、72 h和5、7、14天,各时间点10只。均于第1天开始灌胃持续至实验结束,对照组和模型组灌胃给予生理盐水,珍宝丸组灌胃给予珍宝丸;第7天将乳鼠置于8%O2+92%N2(V/V)的缺氧箱内2 h建立新生鼠脑缺氧模型,分别于缺氧后0.5、24、48、72 h和5、7、14天处死动物取脑组织待测;采用HE染色检测乳鼠脑组织脑室的病理改变。结果对照组各时间点均可见脑室周围神经细胞排列整齐、紧密、逐渐地成熟饱满。与对照组比较,模型组可见随着时间点0.5、24、48、72 h脑室周围神经细胞水肿逐渐的加重,细胞排列松散,伴随脑缺氧的脑细胞内外水肿明显,至72 h达高峰;5、7、14天随着时间点的延续逐渐缓解,至14天明显缓解。珍宝丸组可见随着时间点0.5、24、48、72 h神经细胞水肿逐渐的加重,细胞排列松散,伴随脑缺氧的脑细胞内外水肿较明显,至72h达高峰,各时间点病变均较模型组轻;5、7、14天随着时间点的延续逐渐缓解,至5天明显缓解,7、14天与对照组无明显差异。结论从病理改变可知,珍宝丸预处理可减轻各时间点缺氧对乳鼠脑室的损伤,从而对缺氧性脑损伤具有明确的保护作用。
Objective To observe the protective effect of Zhenbao Pill pretreatment on cerebral ventricle in neonatal hypoxic neonatal rats at different time points. Methods One hundred and twenty fresh Wistar rats were randomly divided into control group, model group and Zhenbao pill group. Each group was divided into seven time points: 0.5, 24, 48, 72 h and 5, 7, 14 days, 10 at each time point. The rats in the control group and the model group were intragastrically given normal saline and the Zhenbao pill group were intragastrically administered with Zhenbao pill. On the 7th day, the suckling mice were placed in 8% O2 + 92% N2 (V / V) hypoxia for 2 hours to establish neonatal rat model of hypoxia, were sacrificed at 0.5,24,48,72 h and 5,7,14 days after hypoxia, respectively. The brain tissues were harvested and tested by HE staining Neonatal rat brain tissue pathological changes. Results The control group at each time point can be seen around the periventricular nerve cells arranged in neat, close, gradually mature and full. Compared with the control group, in the model group, the edema of peripheral nerve cells at 0.5, 24, 48 and 72 h was gradually aggravated, and the cells were loosely arranged. The edema of brain cells with hypoxia was obvious and peaked at 72 h. 5, 7, 14 days gradually relieved with the extension of time point, obviously relieved to 14 days. The group of Zhenbao pill shows that the edema of neurons gradually increases at 0.5,24,48,72 h, the cells are loosely arranged, the edema of brain cells with hypoxia is obvious, reaching the peak at 72h, and the lesions at all time points are more serious Model group was light; 5, 7, 14 days gradually relieved with the extension of time point, obviously relieved to 5 days, and there was no significant difference between 7 and 14 days. Conclusion From the pathological changes, it can be seen that Zhenbao Pill pretreatment can reduce the damage of neonatal rat ventricle at various time points and thus have a clear protective effect on hypoxic brain injury.
