本文用3H-TdR掺入试验发现空肠弯曲菌43KD热休克蛋白(HSP43)能显著促使小鼠脾淋巴细胞活化,并呈剂量依赖关系。这种活化作用不但能被特异性针对HSP43的二株单抗明显抑制,而且也能被针对HSP60家族保守区序列的二株单抗所抑制。去除巨噬细胞同样也能抑制HSP43活化作用。用单抗和补体的特异性杀伤所致T细胞亚群缺失试验发现HSP43主要活化CD4+T细胞。本文结果提示空肠弯曲菌HSP43可能具有某些超抗原特性,这对解释HSP诱导自身免疫病具有一定意义。
In this paper, 3H-TdR incorporation assay showed that 43KD heat shock protein (HSP43) of Campylobacter jejuni could significantly promote the activation of spleen lymphocytes in a dose-dependent manner. This activation can not only be significantly inhibited by the two monoclonal antibodies specific for HSP43, but also by the two monoclonal antibodies against the conserved region of the HSP60 family. Removal of macrophages also inhibits HSP43 activation. T cell subpopulation depletion induced by specific killing of McAb and complement was found to activate CD4 + T cells primarily. Our results suggest that Campylobacter jejuni HSP43 may have some superantigen characteristics, which is of some significance for the explanation of HSP-induced autoimmune diseases.
