目的: 建立人血浆中己酮可可碱浓度测定法。方法:采用反相高效液相色谱法。结果: 方法准确度为826 % ~1059 % (25 ~300 ng/ ml) ;批内 R S D 分别为76 % (50 ng/ml) 、58 % (100 ng/ml) 和39 % (150 ng/ml) ,批间 R S D 分别为108 % (50 ng/ ml) 、79 % (100 ng/ ml) 和41 % (150 ng/ ml) ;最低检测浓度为25 ng/ ml( S/ N= 3) 。结论:本法准确、可靠、灵敏,适用于己酮可可碱新剂型的生物利用度和药代动力学研究。
Objective: To establish a method for the determination of pentoxifylline in human plasma. Methods: Reversed-phase high performance liquid chromatography. Results The accuracy of the method was 826% -1059% (25 ~ 300 ng / ml). The intra-operative R SD was 76% (50 ng / ml) and 58% (100 ng / ml) and 39% (150 ng / ml) respectively. The inter-assay RSD was 108% (50 ng / ml), 79% (100 ng / ml) and 41% / Ml); the lowest detection concentration was 25 ng / ml (S / N = 3). Conclusion: This method is accurate, reliable, sensitive and suitable for the study of bioavailability and pharmacokinetics of pentoxifylline.
