目的探讨补肾活络方治疗多囊卵巢综合征(PCOS)的药理机制。方法 60只雌性SD大鼠分为空白组、模型组、阳性药物组、补肾活络方高、中、低剂量组,每组10只。采用胰岛素联合绒毛膜促性腺激素(HCG)造成的多囊卵巢(PCO)大鼠模型,以补肾活络方高、中、低3个剂量以及达英-35分别作用于PCO模型大鼠12 d后,放免法检测动物血清促黄体生成素(LH)、睾酮(T)、促卵泡生成素(FSH)、雌二醇(E2)的含量,光镜下观察各组大鼠卵巢形态。结果与模型组相比,补肾活络方高、中剂量组大鼠卵巢重量显著减轻(P<0.05),卵泡数目显著减少(P<0.05),组织形态明显改善,血清T、LH含量显著降低(P<0.05)。结论补肾活络方可能通过降低T、LH的水平而达到促进卵泡发育和排卵的作用。
Objective To investigate the pharmacological mechanism of Bushen Huolixifang in the treatment of polycystic ovary syndrome (PCOS). Methods Sixty female SD rats were divided into blank group, model group, positive drug group and Bushen Huoxue Fang high, medium and low dose group, 10 rats in each group. Polycystic ovary (PCO) rat model induced by insulin combined with chorionic gonadotropin (HCG) was treated with high, medium and low doses of Bushen Huoxue Fang and up to -35 in PCO model rats for 12 days The serum levels of luteinizing hormone (LH), testosterone (T), follicle stimulating hormone (FSH) and estradiol (E2) were detected by radioimmunoassay. The morphological changes of ovaries were observed under light microscope. Results Compared with model group, the weight of ovary of rats in Bushen Huoxue Fang group was significantly reduced (P <0.05), the number of follicles was significantly decreased (P <0.05), the morphology of tissues was significantly improved, the contents of serum T and LH were significantly decreased P <0.05). Conclusion Bushen Huoluofang can reduce the level of T and LH to promote follicular development and ovulation.
