目的探讨先天性肾盂输尿管连接部梗阻(PUJO)患儿肾盂输尿管连接部组织中分化抑制因子2(Id2)mRNA的表达情况。方法收集20例接受肾盂成形术的PUJO患儿的肾盂输尿管连接部组织,10例对照组标本来自肾母细胞瘤患儿无肿瘤转移的肾盂输尿管组织。按照部位不同将PUJO组分为扩张肾盂、狭窄段及狭窄段下正常输尿管;对照组分为肾盂和输尿管。用半定量反转录-PCR检测Id2 mRNA在不同组间表达水平。结果 PUJO组:Id2 mRNA在扩张肾盂、狭窄段输尿管间表达差异无统计学意义(P>0.05),而此2组与狭窄段下正常输尿管比较均有降低(Pa<0.05);对照组:肾盂和输尿管Id2 mRNA表达差异无统计学意义(P>0.05);PUJO组扩张肾盂、狭窄段输尿管中Id2 mRNA的表达与对照组中肾盂、输尿管表达比较均显著降低(Pa<0.05),而PUJO组狭窄段下正常输尿管Id2 mRNA表达与对照组肾盂和输尿管比较差异均无统计学意义(Pa>0.05)。结论 Id2基因可能在PUJO的发病机制中发挥重要作用。
Objective To investigate the expression of Differentiation Inhibitor 2 (Id2) mRNA in the ureteropelvic junction of children with congenital ureteropelvic junction obstruction (PUJO). METHODS: Twenty ureteropelvic junctional tissues of 20 PUJO children undergoing pyeloplasty were collected, and 10 control samples were obtained from the ureteropelvic ureter without tumor metastasis in nephroblastoma. According to the different parts of the PUJO group is divided into dilated renal pelvis, stenosis and stricture section of the normal ureter; control group is the renal pelvis and ureter. The expression of Id2 mRNA in different groups was detected by semi-quantitative reverse transcription-PCR. Results In the PUJO group, there was no significant difference in the expression of Id2 mRNA between the expanded renal pelvis and the narrow ureter (P> 0.05), but both of the two groups were significantly lower than those in the normal ureter (P <0.05). In the control group, (P> 0.05). The expression of Id2 mRNA in ureter and ureter in the PUJO group was significantly lower than that in the control group (P <0.05), while the expression of Id2 mRNA in PUJO group was significantly lower than that in the PUJO group There was no significant difference in the expression of Id2 mRNA between normal ureter and ureter in the control group (P> 0.05). Conclusion Id2 gene may play an important role in the pathogenesis of PUJO.
