目的:研究白介素-8(IL-8)能否促成人卵巢癌SKOV3细胞系细胞肿瘤干细胞样细胞特性。方法:用添加或未添加重组人IL-8(rh IL-8:10.0 ng/m L)的干细胞条件培养基处理SKOV3细胞。肿瘤球形成和琼脂集落形成试验测定球形成率和集落形成率。蛋白质印迹分析肿瘤干细胞标志蛋白CD133和CD44表达水平。结果:rh IL-8(10.0 ng/m L)显著地增高SKOV3细胞肿瘤球形成率和琼脂集落形成率。此外,rh IL-8(10.0 ng/m L)上调SKOV3细胞CD133和CD44蛋白表达。结论:rh IL-8具有促进SKOV3细胞肿瘤干细胞样细胞特性作用。
Objective: To investigate whether interleukin-8 (IL-8) can promote the tumor cell characteristics of SKOV3 cells in human ovarian cancer cells. Methods: SKOV3 cells were treated with stem cell conditioned media with or without recombinant human IL-8 (rh IL-8: 10.0 ng / ml). Tumor formation and agar colony formation assays determine the rate of bulb formation and colony formation. Western blot analysis of tumor stem cell markers CD133 and CD44 expression levels. Results: rh IL-8 (10.0 ng / m L) significantly increased the rate of tumor formation and the rate of colony formation in SKOV3 cells. In addition, rh IL-8 (10.0 ng / m L) up-regulated CD133 and CD44 protein expression in SKOV3 cells. Conclusion: rh IL-8 can promote tumor stem cell-like cell characteristics of SKOV3 cells.
