目的 建立适用于体外药理学研究的破骨细胞骨吸收功能的定量评定方法。方法 机械分离法获得破骨细胞 ,接种于象牙簿片底物上 ,分别于培养 1d、3d、5 d、7d取出象牙簿片各 4张 ,采用光镜和扫描电镜观察骨吸收陷窝数量的变化 ;对随机拍摄的陷窝照片进行计算机形态计量分析 ,测算陷窝的面积和深度。结果 骨吸收陷窝计数光镜法和扫描电镜法具有良好的相关性 ,r=0 .9998,P<0 .0 0 1。随着培养时间的延长 ,陷窝数量逐渐增多、面积扩大、深度加深。结论 骨吸收陷窝计数光镜法可以用于体外培养破骨细胞功能的定量评价 ,结合陷窝面积和深度分析 ,可以较为全面地评价体外培养破骨细胞的骨吸收功能
Objective To establish a quantitative assessment method of osteoclast bone resorption in vitro pharmacology. Methods Osteoclasts were obtained by mechanical separation method and inoculated on the surface of ivory tablets. Four slices of ivory tablets were taken out on day 1, day 3, day 5, day 7 respectively. The number of bone resorption lacuna was observed by light microscope and scanning electron microscope Change; the random photograph of the pit photo computer morphometry analysis, calculation of lacunae area and depth. Results The bone resorption lacuna count light microscopy and scanning electron microscopy had a good correlation, r = 0.9998, P <0.001. With the extension of incubation time, the number of dimples gradually increased, the area expanded and the depth deepened. Conclusion The bone resorption lacuna counting light microscopy can be used to evaluate the function of osteoclasts cultured in vitro. Combined with the size and depth of dimple, the bone resorption function of cultured osteoclasts can be evaluated more fully
