目的探讨系统性红斑狼疮(SLE)患者外周血单个核细胞中MBD4 mRNA表达及其与疾病发病机制、疾病活动性的关系。方法以β-actin为内参照,建立逆转录实时荧光定量聚合酶链反应(FQ-RT-PCR)方法,在Light CyclerReal time PCR检测仪上检测70例SLE患者(活动期32例、缓解期38例)、其他结缔组织病(CTD)患者30例(疾病对照组)、健康体检者30例(正常对照组)外周血单个核细胞(PBMCs)中MBD4 mRNA的表达水平和含量。以ΔCt=Ct(待测基因)-Ct(内参基因)来比较基因表达水平的高低。结果 SLE患者PBMC中MBD4 mRNA表达水平高于正常对照组与疾病对照组(P<0.05),且疾病不同病期(活动期与缓解期)MBD4 mRNA表达水平差别有统计学意义(P<0.01)。结论结果显示SLE患者PBMC中MBD4 mRNA表达水平显著高于正常人、疾病对照组,提示MBD4的表达与SLE的发生发展存在一定关联性,可能参与了SLE的发病过程并与疾病的活动性有关。
Objective To investigate the expression of MBD4 mRNA in peripheral blood mononuclear cells of patients with systemic lupus erythematosus (SLE) and its relationship with pathogenesis and disease activity. Methods β-actin was used as an internal reference to establish reverse transcription real-time quantitative polymerase chain reaction (FQ-RT-PCR). 70 patients with SLE (active stage 32, remission 38 (Control group), 30 cases of other connective tissue disease (CTD) patients (control group) and 30 healthy control subjects (normal control group), the level of MBD4 mRNA in peripheral blood mononuclear cells (PBMCs) The level of gene expression was compared with ΔCt = Ct (gene to be tested) -Ct (internal reference gene). Results The expression of MBD4 mRNA in peripheral blood mononuclear cells of SLE patients was higher than that of normal controls and controls (P <0.05), and the expression of MBD4 mRNA in different disease stages (active and remission) was significantly different (P <0.01) . Conclusion The results showed that the expression of MBD4 mRNA in peripheral blood mononuclear cells of SLE patients was significantly higher than that of controls and normal controls, suggesting that the expression of MBD4 correlated with the occurrence and development of SLE. It may be involved in the pathogenesis of SLE and related to the activity of the disease.
