研究海洋放线菌WB-F5发酵液的抑菌活性,并分离纯化其抑菌作用物。通过琼脂打孔法测定WB-F5发酵液对各菌株的抑制活性。本实验选用大孔树脂吸附法、硅胶柱层析法等方法,分离纯化活性物质。WB-F5发酵液对金黄色葡萄球菌、耐甲氧西林金黄色葡萄球菌(MRSA)、枯草杆菌等革兰氏阳性菌有较好的抑制作用,同时确定大孔吸附树脂AB-8和硅胶柱层析作为分离手段较为适宜,并且得到化合物WBF5-F,经结构解析证明其为5,7,4′-三羟基异黄酮,即染料木素。WB-F5是一株能产生高效抗菌活性物质的海洋放线菌,对WB-F5及其代谢产物进行研究,将有助于抗生素药物的开发研究。
The antibacterial activity of marine actinomycete WB-F5 fermentation broth was studied, and its antibacterial activity was isolated and purified. The inhibitory activity of WB-F5 fermentation broth on each strain was determined by agarose drilling. The experiment selected macroporous resin adsorption method, silica gel column chromatography and other methods, separation and purification of active substances. WB-F5 fermentation broth against Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), Bacillus subtilis and other Gram-positive bacteria have a good inhibitory effect, while identifying the macroporous resin AB-8 and silica gel column Chromatography is more suitable as a separation method, and the compound WBF5-F is obtained, which is proved to be 5,7,4’-trihydroxyisoflavone, namely genistein, by structural analysis. WB-F5 is a marine actinomycete that produces highly effective antimicrobial active substances. The study of WB-F5 and its metabolites will be helpful for the development of antibiotic drugs.
