目的:研究博落回提取物中血根碱(Sanguinarine,SAN)对舌癌体外增殖、凋亡和迁移作用的影响及其机制。方法:采用不同浓度的血根碱处理Tscca和Tca8113细胞24、48、72 h,MTT法检测血根碱对两种舌癌细胞增殖抑制的影响;用药48 h后,流式细胞仪Annexin V-FITC/PI染色法检测不同浓度血根碱对细胞凋亡的影响;Western blotting法检测不同浓度血根碱对细胞中NF-κB表达的影响;细胞划痕实验检测不同浓度血根碱对舌癌细胞迁移能力的影响。结果:经血根碱处理后的舌癌细胞系Tscca和Tca8113有明显的抑制增殖作用,并呈现一定的浓度和时间依赖关系。血根碱作用于舌鳞癌Tca8113细胞24 h、48 h、72h的半数抑制浓度(IC50)分别为2.53μmol/L、2.38μmol/L、1.96μmol/L。舌鳞癌Tscca细胞24 h、48 h、72h的半数抑制浓度(IC_(50))分别为1.48μmol/L、1.17μmol/L、0.89μmol/L。血根碱可以抑制NF-κB的表达,且有一定的剂量相关性。一定浓度的血根碱还能抑制Tca8113细胞株的迁移。结论:血根碱能显著抑制舌癌细胞Tscca和Tca8113的增殖并且诱导其凋亡,其机制可能与对NF-κB表达的影响有关,同时血根碱可影响Tca8113细胞株的迁移。
Objective: To study the effect and mechanism of Sanguinarine (SAN) on the proliferation, apoptosis and migration of tongue cancer cells in vitro. Methods: TcaC and Tca8113 cells were treated with different concentrations of sanguinarine for 24,48,72 h. MTT assay was used to detect the effect of sanguinarrhizae on proliferation inhibition of two tongue cancer cells. Flow cytometry Annexin V- FITC / PI staining was used to detect the effect of different concentrations of hematoxylin on cell apoptosis; Western blotting was used to detect the effect of different concentrations of hematoxylin on the expression of NF-κB; Cell scratch assay was used to detect the effect of different concentrations of hematoxylin on tongue cancer Effect of cell migration ability. Results: The tongue squamous carcinoma cell lines Tscca and Tca8113 had significant inhibitory effects on proliferation, and showed a certain concentration and time-dependent relationship. The IC50 values of sanguinarine on Tca8113 cells were 2.53μmol / L, 2.38μmol / L and 1.96μmol / L at 24 h, 48 h and 72 h respectively. The median inhibitory concentration (IC 50) of TcaSCs in tongue squamous cell carcinoma at 24 h, 48 h and 72 h were 1.48 μmol / L, 1.17 μmol / L and 0.89 μmol / L, respectively. Scumerine can inhibit the expression of NF-κB, and have a certain dose-related. A certain concentration of sanguinarine can also inhibit Tca8113 cell line migration. Conclusion: Scualate can significantly inhibit the proliferation and induce the apoptosis of Tcaca cells and Tca8113 cells, which may be related to the effect on the expression of NF-κB. Meanwhile, sage base can affect the migration of Tca8113 cells.
