目的探讨人乳头状瘤病毒(human papillomavirus,HPV)16和(或)18 E6蛋白在宫颈脱落细胞中的表达与宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)及宫颈鳞状细胞癌(cervical squamous cell carcino-ma,CSCC)的关系。方法采用免疫印迹(Western blot)方法检测2003年6月至2005年6月中国医科大学第一附属医院CSCC30例、CIN52例和16例正常宫颈脱落细胞中HPV16和(或)18E6蛋白的表达。结果在上述各组宫颈脱落细胞中HPV16和(或)18E6表达阳性率分别76.7%(23/30)、51.9%(27/52)和12.5%(2/16);其中在CSCC宫颈脱落细胞中HPV16和(或)18E6明显高于正常对照组的表达(P<0.01);CINⅡ、Ⅲ组的表达也显著高于对照组(P<0.05),而CINⅠ组与正常对照组比较差异无显著性意义(P>0.05)。结论宫颈脱落细胞标本存在HPV16和(或)18 E6癌基因的表达。宫颈脱落细胞中HPV16和(或)18 E6癌基因的过度表达与CSCC及CINⅡ、Ⅲ密切相关。
Objective To investigate the expression of human papillomavirus (HPV) 16 and / or 18 E6 protein in cervical exfoliated cells and their relationship with cervical intraepithelial neoplasia (CIN) and cervical squamous cell carcino-ma, CSCC). Methods Western blot was used to detect the expression of HPV16 and / or 18E6 protein in 30 cases of CSCC, 52 cases of CIN and 16 cases of normal cervical exfoliated cells from June 2003 to June 2005 in the First Affiliated Hospital of China Medical University. Results The positive expression rates of HPV16 and / or 18E6 in cervical exfoliated cells were 76.7% (23/30), 51.9% (27/52) and 12.5% (2/16), respectively. Among them, The expression of HPV16 and / or 18E6 was significantly higher than that of the normal control group (P <0.01); the expression of CINⅡ and Ⅲ was also significantly higher than that of the control group (P <0.05), while there was no significant difference between the CINⅠgroup and the normal control group Significance (P> 0.05). Conclusion The expression of HPV16 and / or 18 E6 oncogene exists in exfoliated cells of cervical specimens. Overexpression of HPV16 and / or E6 oncogene in cervical exfoliated cells is closely related to CSCC and CINII, III.
