目的:采用单因素考察和正交试验设计,对复方中药柴胡疏肝散提取工艺进行优化。方法:采用RP-HPLC法测定柴胡疏肝散中质量控制指标柴胡皂苷a、阿魏酸、橙皮苷、芍药苷的含量。以4个活性成分的含量测定结果与浸膏得率为考察指标,选用L9(34)正交表安排实验,对乙醇浓度、乙醇用量、回流时间、回流次数4个因素进行优化。结果:柴胡疏肝散最佳提取工艺为加入10倍量体积分数为80%的乙醇,回流提取2次,每次90 min。结论:本实验为柴胡疏肝散的制剂开发提供了实验依据。
OBJECTIVE: To optimize the extraction process of compound traditional Chinese medicine Bupleurum Shugan Powder by single factor and orthogonal test design. Methods: The quality control indexes of saikosaponin a, ferulic acid, hesperidin and paeoniflorin in Radix Bupleuri Shugan Powder were determined by RP-HPLC. The determination results of the content of the four active ingredients and the extraction rate of the extract were used as indicators. The L9 (34) orthogonal table was used to arrange the experiment. The ethanol concentration, the amount of ethanol, the reflux time, and the number of reflux were optimized. Results: The best extraction process of Radix Bupleuri Shugan San was to add 10 times the volume fraction of 80% ethanol, and reflux extraction for 2 times, 90 min each time. Conclusion: This experiment provides experimental basis for the development of preparations of Chaihu Shugan Powder.
