目的应用生物信息学预测分析人乳头瘤16型病毒(human papillomavirus type 16,HPV16)E7蛋白的结构特征与抗原表位。方法自NCBI GENE BANK获取HPV16E7蛋白的基因信息,应用ProtParam分析其编码的E7蛋白理化性质和一级结构;利用SOPMA分析E7蛋白的二级结构;应用SWISS MODEL的Automated mode预测E7蛋白的三维空间结构;使用NetNGlyc 1.0Server、NetPhos3.1Server、MotifyScan、SignalP及TMHMM预测蛋白质的翻译后修饰位点、信号肽切割位点和跨膜区;利用CLUSTALX将HPV16E7蛋白同其他高危型HPV E7蛋白进行同源性对比;应用ABCpred、BepiPred、IEDB、SYFPEITHI和NetMHCIIpan 3.1Server预测E7蛋白的优势抗原表位。结果生物信息学预测E7蛋白无规卷曲占52.04%,结构较松散,为不稳定性蛋白。该蛋白存在多个磷酸化位点。利用序列匹配度为46%的HPV45E7蛋白为模板得到同型二聚体三级预测结构。E7蛋白高度保守,且具有潜在的B细胞抗原表位1-8、15-22、33-46,CD8+T细胞抗原表位82-90、11-19、7-11、85-93、13-21和CD4+T细胞抗原表位9-23、73-87、10-24、7-21、21-35。结论 HPV16E7蛋白为不稳定蛋白。该蛋白高度保守,且具有潜在的B、T细胞抗原表位。
Objective To predict the structural characteristics and antigenic epitopes of E7 protein of human papillomavirus type 16 (HPV16) using bioinformatics. Methods The gene information of HPV16E7 protein was obtained from NCBI GENE BANK. The physical and chemical properties and the primary structure of E7 protein were analyzed by ProtParam. The secondary structure of E7 protein was analyzed by SOPMA. The 3D structure of E7 protein was predicted by using Automated mode of SWISS MODEL ; Using NetNGlyc 1.0Server, NetPhos3.1Server, MotifyScan, SignalP and TMHMM to predict the post-translational modification sites, signal peptide cleavage sites and the transmembrane region of the protein; using CLUSTALX to homology HPV16E7 protein with other high-risk HPV E7 proteins The predominant epitopes of E7 protein were predicted by ABCpred, BepiPred, IEDB, SYFPEITHI and NetMHCIIpan 3.1Server. Results Bioinformatics predicted that the random coil of E7 protein accounted for 52.04%, with loose structure and unstable protein. The protein has multiple phosphorylation sites. Using the HPV45E7 protein with a sequence match of 46% as a template, a tertiary homodimer prediction structure was obtained. The E7 protein is highly conserved and has potential B cell epitopes 1-8, 15-22, 33-46, CD8 + T cell epitopes 82-90, 11-19, 7-11, 85-93, -21 and CD4 + T cell epitopes 9-23, 73-87, 10-24, 7-21, 21-35. Conclusion HPV16E7 protein is unstable protein. This protein is highly conserved and has potential B and T cell epitopes.
