目的:建立HPLC法测定炮制后大蓟中柳穿鱼苷元的含量,并以其含量为指标,优选大蓟最佳炮制方法。方法:色谱柱:Hypersil ODS2柱(4.6 mm×250 mm,5μm);检测波长:330 nm;流动相:甲醇-水(55:45);柱温:45℃;流速:1.0 mL/min;进样量:10μL;以柳穿鱼苷元的含量为指标,以大蓟炮制的温度和时间作为影响因素,对大蓟的炮制工艺进行优选。结果:柳穿鱼苷元在4~400μg/mL(r=0.9998,n=6)的范围内线性关系良好,平均回收率(n=6)为99.03%,RSD为1.62%。确定大蓟最佳炮制工艺为190℃,烘制10 min。结论:该法简便快速,重复性好,准确度高,为大蓟的合理炮制提供参考。
OBJECTIVE: To establish a HPLC method for the determination of the content of perisinone in Salix sinensis after processing, and to use its content as an index, to optimize the best preparation method of Radon. Method: Column: Hypersil ODS2 column (4.6 mm×250 mm, 5 μm); detection wavelength: 330 nm; mobile phase: methanol-water (55:45); column temperature: 45°C; flow rate: 1.0 mL/min; Sample volume: 10 μL; the content of Salviol was used as an index, and the temperature and time of concocting were used as the influencing factors to optimize the concocting process. Results: In the range of 4-400 μg/mL (r=0.9998, n=6), there was a good linear relationship between the tiopianin and the average recovery (n=6) was 99.03%, and the RSD was 1.62%. It was determined that the best processing technology of Dalat was 190°C and baked for 10 minutes. Conclusion: The method is simple, rapid, reproducible and accurate.
