目的研究乐胃饮加味方对SGC-7901人胃癌细胞株核因子-κB(nuclear factor kappa B,NF-κB)和信号传导与转录激活因子(signal transducers and activators of transcription 3,STAT3)相互作用的影响。方法以SGC-7901人胃癌细胞株为研究对象,构建NF-κB(Rel A/p65)-PECFP和STAT3-PEYFP载体,转染至SGC-7 9 0 1人胃癌细胞,LPS刺激后,应用乐胃饮加味方干预。应用荧光共振能量转移技术和免疫共沉淀技术检测干预后NF-κB和STAT3蛋白在细胞中的交互作用。结果经LPS刺激后,NF-κB和STAT3的相互作用增强,用乐胃饮加味方干预后,NF-κB和STAT3的相互作用明显减弱。结论乐胃饮加味方可能通过阻断NF-κB和STAT3蛋白间的交互作用,可用于治疗胃癌前病变,预防胃癌生成。
Objective To study the effect of Lewei Yin Jiawei Recipe on the interaction between nuclear factor-kappa B (NF-κB) and signal transducers and activators of transcription 3 (STAT3) in human gastric cancer cell line SGC-7901 influences. Methods SGC-7901 human gastric cancer cell line was used as the research object to construct NF-κB (Rel A / p65) -PECFP and STAT3-PEYFP vector and transfected into SGC-7901 gastric cancer cells. After LPS stimulation, Stomach drink flavored square intervention. Fluorescence resonance energy transfer and co-immunoprecipitation were used to detect the interaction of NF-κB and STAT3 in the cells after intervention. Results After stimulation with LPS, the interaction between NF-κB and STAT3 was enhanced. The interaction between NF-κB and STAT3 was significantly weakened after intervention with LEGO recipe. Conclusion Lewei Yin Decoction can prevent gastric cancer by treating the interaction between NF-κB and STAT3 protein and treating gastric precancerous lesions.
