内皮细胞衬里于人工血管使人工血管内皮化,从而使人工血管具有抗血小板粘附聚集的功能。为小口径人工血管的应用开辟了新的途径,国外已成功地应用于临床,国内亦已进行灵长类动物的研究并获得满意效果。目前主要采用多普勒超声及血管造影检查,间接反应内皮化人工血管抗血小板粘附聚集的功能。本实验首次用~(99m)Tc标记恒河猴血小板,对内皮化人工血管内皮细胞抗血小板粘附聚集功能进行了体外和恒河猴体内的研究。 首先,建立了同素~(99m)Tc标记恒河猴的血小板方法。采用10例雄性恒河猴静脉血,分离血小板后进行体外标记,以亚锡离子作游离~(99m)Tc的还原剂,分析了亚锡离子浓度和亚锡离子与体外血小板作用不同时间对血小板标记率的影响,鉴定丁标记血小板的放化纯度及其稳定性,比较了血小板标记前、后聚集功能的变化。证实在4.5mmol/L亚锡离子浓度、亚锡离子与血小板作用30分钟的条件下,血小板标记率最高,本方法所得标记血小板放化纯度高、性质稳定,血小板标记后血小板的聚集的功能未受损害。 其次,应用~(99m)Tc标记血小板进行内皮化人工血管内皮细胞抗血小板粘附聚集功能的体外研究。分三组人工血管,每组各6条,A组为无内皮细胞种植组,B组为内皮细胞种植后孵育2小时,C组为内皮细胞种植后孵育9天。在体外模拟循?
