目的:观察益肾降浊方对5/6肾切除慢性肾衰(CRF)大鼠肾组织TGF-β_1、BMP-7及体外培养SD大鼠肾小球系膜细胞(MC)和肾间质成纤维细胞(FC)增殖情况,探讨益肾降浊方治疗慢性肾功能衰竭的机理。方法:采用动物模型和细胞培养的方法。清洁级7周龄雄性SD大鼠60只,随机分为空白组、假手术组、CRF模型组、CRF对照组、CRF治疗组。分别进行手术造模并予不同药物治疗后,免疫组化法观察肾组织TGF-β_1、BMP-7;体外培养SD大鼠肾小球系膜细胞(GMC)、肾间质成纤维细胞(RIF),制备各组含药血清,细胞给药后MTT法测定细胞增殖情况。结果:益肾降浊方可降低CRF大鼠血清BUN、SCr(P<0.05),降低残余肾组织中TGF-β_1而提高BMP-7(P<0.05);肾组织HE染色结果示肾组织纤维化减轻;益肾降浊方含药血清能抑制体外培养SD大鼠GMC和RIF的增殖(P<0.05)。结论:益肾降浊方能够有效地减缓慢性肾衰肾组织纤维化发展,是防治慢性肾衰的有效方药。
Objective: To observe the effect of Yishen Jiangzhuo decoction on the expression of TGF-β 1, BMP-7 in renal tissue and renal tubulointerstitial (MC) in 5/6 nephrectomized rats with chronic renal failure (CRF) Fibroblasts (FC) proliferation, explore the mechanism of Yishen Jiangzhuo Fang in the treatment of chronic renal failure. Methods: Animal models and cell culture methods were used. Sixty male Sprague-Dawley rats were randomly divided into blank group, sham operation group, CRF model group, CRF control group and CRF treatment group. The rats were randomly divided into three groups: the model group and the model group. The rats were divided into three groups: ), The drug-containing serum of each group was prepared, and the cell proliferation was measured by MTT method after the cells were administered. Results: Yishenjiangzhuo decoction could reduce BUN, SCr (P <0.05), reduce TGF-β1 in residual renal tissue and increase BMP-7 (P <0.05); renal tissue HE staining showed that renal tissue fibers Yishen Jiangzhuo containing serum can inhibit the proliferation of GMC and RIF in SD rats cultured in vitro (P <0.05). Conclusion: Yishen Jiangzhuo Fang can effectively slow the development of renal fibrosis in patients with chronic renal failure and is an effective prescription for prevention and treatment of chronic renal failure.
