利用逆转录聚合酶链反应(RT-PCR)定量检测了10例正常人前列腺、15例前列腺癌(PCa)及 PCa细胞系PC-3m中白细胞介素6受体(IL-6R)基因表达水平。结果发现,正常人前列腺组织中IL-6R基因呈低水平表达,PCa组织中IL-6R基因表达增强,明显高于正常组(P<0.01),且73.3%(11/15)组织样本中IL-6R mRNA表达水平高于 PC-3m。而IL-6所介导的假单胞菌外毒素融合蛋白对该细胞系有强烈的杀伤作用。这表明IL-6及IL-6R与PCa的发生发展有关,并为PCa的生物学治疗的研究奠定了基础。
Quantitative detection of interleukin-6 receptor (IL-6R) gene expression in prostate, 15 PCa, and PC-3m PC-3m cells was performed using reverse transcription polymerase chain reaction (RT-PCR). . The results showed that IL-6R gene expression was low in normal human prostate tissue, and IL-6R gene expression in PCa tissue was enhanced, which was significantly higher than that in the normal group (P<0.01), and IL in 73.3% (11/15) tissue samples. The expression level of -6R mRNA was higher than that of PC-3m. The IL-6-mediated Pseudomonas exotoxin fusion protein has a strong killing effect on the cell line. This shows that IL-6 and IL-6R are related to the occurrence and development of PCa, and laid the foundation for the study of the biological treatment of PCa.
