目的:检测类肿瘤干细胞(cancer stem-like cell,CSC)标记物(CD44、ALDH1、NANOG)及缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)在口腔鳞癌组织中的表达并进行相关性分析。方法:应用免疫组化法检测40例重庆医科大学附属口腔医院颌面外科手术切除的口腔鳞癌组织、10例癌旁组织、10例淋巴结转移的淋巴结组织以及10例正常淋巴结组织中CD44、ALDH1、NANOG及HIF-1α的表达,采用Fisher确切概率法及秩和检验分析这些标记物与HIF-1α之间及其与肿瘤分化程度、分期及淋巴结转移的关系。结果:(1)CD44、NANOG、HIF-1α在口腔鳞癌组织与癌旁组织中的表达差异均存在统计学意义(P_(CD44)=0.000;P_(NANOG)=0.000;P_(HIF-1α)=0.000);(2)HIF-1α与CD44在口腔鳞癌中的表达存在相关性(r_s=0.498,P=0.027);(3)CD44、ALDH1、NANOG及HIF-1α在口腔鳞癌组织中的表达与分化程度、临床分期及淋巴结转移均无明显相关性。结论:CD44、NANOG、HIF-1α在口腔鳞癌组织中呈高表达,其中CD44与HIF-1α的表达具有相关性。联合检测CD44、NANOG、HIF-1α可能对口腔鳞癌早期诊断具有重要意义。
OBJECTIVE: To detect the expressions of CD44, ALDH1, NANOG and hypoxia inducible factor-1α (HIF-1α) in oral squamous cell carcinoma And correlation analysis. Methods: Immunohistochemical method was used to detect the expression of CD44, ALDH1 in 40 cases of oral squamous cell carcinoma, 10 cases of paracancerous tissues, 10 cases of lymph node metastasis and 10 cases of normal lymph nodes excised from maxillofacial surgery in Affiliated Stomatological Hospital of Chongqing Medical University. , NANOG and HIF-1α were detected by Fisher’s exact test and rank sum test. The relationship between these markers and HIF-1α and the degree of tumor differentiation, stage and lymph node metastasis were analyzed. Results: (1) The expressions of CD44, NANOG and HIF-1α in oral squamous cell carcinoma and adjacent normal tissues were significantly different (P_ (CD44) = 0.000; P_ (NANOG) = 0.000; P_ ) = 0.000); (2) There was a correlation between the expression of HIF-1αand CD44 in oral squamous cell carcinoma (r_s = 0.498, P = 0.027); (3) The expression of CD44, ALDH1, NANOG and HIF- There was no significant correlation between the expression and the degree of differentiation, clinical stage and lymph node metastasis. Conclusion: CD44, NANOG and HIF-1α are overexpressed in oral squamous cell carcinoma and there is a correlation between CD44 and HIF-1α expression. Combined detection of CD44, NANOG and HIF-1α may be of great significance in the early diagnosis of oral squamous cell carcinoma.
