目的观察七氟烷麻醉后对大鼠不同脑区、不同时期腺苷酸环化酶及磷酸二酯酶活性的影响,探讨3’,5’-环腺苷酸(cAMP)在七氟烷麻醉中的作用。方法SD大鼠40只,分层随机区组设计分成5组,每组8只,吸入1.5%的七氟烷,分别在未吸入七氟烷时(对照组)、翻正反射即将消失时(诱导期组)、翻正反射消失后2min(麻醉期组)、翻正反射恢复即刻(恢复期组)和翻正反射恢复后1h(清醒期组)断头取脑。放射免疫法测定大脑皮质、海马和脑干的腺苷酸环化酶(AC)活性及磷酸二酯酶(PDE)活性。结果(1)在对AC活性影响上,与对照组比较,诱导期组、麻醉期组的皮质、海马、脑干及恢复期组皮质、海马AC活性升高;与麻醉期组比较,清醒期组皮质、海马、脑干AC活性降低。上述差异均有统计学意义(均P<0.05)。(2)在对PDE活性的影响上,与对照组比较,诱导期组、麻醉期组皮质、海马、脑干PDE活性下降;与麻醉期组比较,恢复期组、清醒期组皮质、海马、脑干PDE活性升高。上述差异均有统计学意义(均P<0.05)。结论cAMP在七氟烷的全麻机制中可能发挥重要作用。
Objective To observe the effects of sevoflurane anesthesia on the activity of adenylate cyclase and phosphodiesterase in different brain regions and different periods in rats and to investigate the effect of cAMP on sevoflurane anesthesia In the role. Methods Forty Sprague-Dawley rats were randomly divided into five groups with 8 rats in each group. 1.5% sevoflurane was inhaled. When sevoflurane was not inhaled (control group) Induction group), 2 minutes after the inversion reflex disappeared (anesthesia group), immediate reflex recovery (recovery group) and 1 h after reflex reflex recovery (awake group) decapitated brain. Radioimmunoassay was used to determine adenylyl cyclase (AC) activity and phosphodiesterase (PDE) activity in cerebral cortex, hippocampus and brain stem. Results (1) Compared with the control group, AC activity in cortex, hippocampus, brainstem and convalescent group in induction group and anesthesia group was higher than that in control group Cortical, hippocampal, brain stem AC activity decreased. The above differences were statistically significant (all P <0.05). (2) Compared with control group, PDE activity in cortex, hippocampus and brainstem of induction group and anesthesia group decreased in PDE activity group. Compared with control group, the PDE activity in cortex, hippocampus, Brain stem PDE activity increased. The above differences were statistically significant (all P <0.05). Conclusion cAMP may play an important role in the general anesthesia mechanism of sevoflurane.
