目的:采用反相高效液相色谱法建立同时测定龟鹿补肾丸中二苯乙烯苷、橙皮苷、朝藿定B和淫羊藿苷含量的方法。方法:采用Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱,流动相为甲醇-水,梯度洗脱(0~10 min,40%A→55%A;10~14min,55%→70%A;14~20 min,70%A);流速:1.0 mL.min-1;检测波长:270 nm;柱温:35℃。结果:二苯乙烯苷、橙皮苷、朝藿定B、淫羊藿苷质量浓度分别在4.20~84.0、3.60~72.0、0.408~8.16、0.552~11.0μg.mL-1范围内呈良好的线性关系(r=0.9991~0.9996);平均回收率(n=6)分别为99.4%、101.0%、99.7%、98.8%,RSD分别为2.7%、1.2%、2.6%和2.7%。结论:本方法适用于龟鹿补肾丸中二苯乙烯苷、橙皮苷、朝藿定B和淫羊藿苷的含量测定,可为龟鹿补肾丸的质量控制或中药配伍等方面提供相关参考。
Objective: To establish a method for simultaneous determination of stilbene glycoside, hesperidin, epimedin B and icariin in Guilu Bushen pill by reversed-phase high performance liquid chromatography. Methods: Diamonsil C18 (250 mm × 4.6 mm, 5 μm) column was used. The mobile phase consisted of methanol and water with gradient elution (0-10 min, 40% A → 55% A, 10-14 min, 55% → 70% A; 14-20 min, 70% A); flow rate: 1.0 mL.min-1; detection wavelength: 270 nm; Results: The concentrations of stilbene glycoside, hesperidin, epimedin B and icariin were linear in the range of 4.20-84.0,3.60-72.0,0.408- 8.16,0.552-11.0μg.mL-1, respectively (N = 6) were 99.4%, 101.0%, 99.7% and 98.8%, respectively, with RSDs of 2.7%, 1.2%, 2.6% and 2.7%, respectively. Conclusion: The method is suitable for the determination of stilbene glucoside, hesperidin, epimedin B and icariin in Guilu Bushen Wan, which can provide reference for the quality control or traditional Chinese medicine compatibility of Guilu Bushen pill. .
